Best Practices to Convert from Online Only to Offline
I'm running low on storage space on my home iMac so I purchased a Samsung USB-C thumb drive that I will move my DropBox to. At the same time, I want to change settings to offline or local storage from where online only where it is currently set. What are the best practices for such a change? Delete all files, then re-download after changing the settings to offline or local? Or something else?

Generally, it wouldn't be possible to move the Dropbox folder to a removable thumb drive. However, an external drive might work, depending on your OS.
Could you clarify what version of Mac OS you're currently running?

Thanks for the info, you might be upgraded to the newer version of the Dropbox desktop application for Mac OS, which prevents having the Dropbox folder outside of your local drive.
