Although it's recommended that the Dropbox folder be located on the same drive as your operating system, it's not required. You can use the option in Dropbox Preferences (Sync tab) to relocate the folder to another location. There's also an Advanced Options link during installation (after you sign-in) that allows you to specify a different location before Dropbox even starts syncing.

I would just caution you not to locate Dropbox on an external drive. While it is possible, data loss can occur under the right circumstances, such as if the Dropbox is started on the computer before the drive is mounted, or if the drive becomes disconnected or powered off while Dropbox is running.

If the links are external, pointing to files within the Dropbox folder, they will likely break.

Keep in mind that Dropbox does not support symlinks. If they're external pointing into Dropbox, it's not much of an issue, but if they're within your Dropbox folder pointing to files outside of Dropbox, they can cause high CPU usage and problems with syncing (not to mention that they don't sync in real-time).
