Based on everything, it sounds like it all works as it should, so I'm a bit unsure as to what you'd like to achieve here.
You mentioned "Do I need it to be set to sync so it's available offline to accomplish this?". Are you referring to your camera uploads folder, or the new folder you wish to create?
When you say that you want this to sync to your desktop, do you mean a different device? What would you like to achieve on your end?

I guess, can I leave the sync setting on my desktop in the Dropbox folder to 'Online-only' and sort it the way I want so it will sync to dropbox.com that way? Thanks for being patient..

Yes, all changes within your Dropbox folder will sync back to Dropbox.com as long as the desktop app is running and syncing on your computer Cloudsoverkorra
