Deleting the photos on the device wont delete them from Dropbox - just make sure they HAVE actually uploaded first 🙂
In terms of the setting you dont need to change anything. The difference is simply a way of saying 'upload only photos from this point forward' OR 'upload everything'. The first one would be used if you, for example, set up a new device and used a backup to set it up and had already put everything on Dropbox.
