Will I lose everything if I delete the file on my hard drive?
I downloaded the app for my laptop and it put a file on the physical hard drive rather than what I thought was a shortcut to the online storage. It's filled my hard drive. If I delete the hard drive file will I lose everything stored on my online profile? It's my work computer and I store a lot of photos I take at work there so thought a shortcut would be useful but I didn't think it'd fill the hard drive!

The Dropbox folder that the app creates locally on the device, is indeed a syncing point between your account online, and said folder.
Therefore, if you take any action there, it will be reflected online. The safest way to go on about it, would be to uninstall the app, and then delete the files you need to delete.
